چکیده

& R R = spin label incorporated in cu-chymotrypsin and trypsin (1). The spectral results also show the power of applying such a method to structural comparison of active sites of new proteases whose threedimensional structures are not yet known. We present here the detailed syntheses and purifications of these spin labels and some of the specific problems encountered in preparing the inhibited sulfonylated enzyme derivatives.

Spin-labeled Sulfonyl Fluorides as Active Site Probes of Protease Structure

a-Chymotrypsin and trypsin were spin-labeled with 14 ortho-, meta-, and para-substituted phenylsulfonyl fluorides. These analogues of tosylfluoride were substituted with either pyrrolidinyl, pyrrolinyl, or piperidinyl nitroxide moieties. A complete description of their synthesis and the enzyme inhibition studies is presented in the following paper (WONG, S. S., QUIGGLE, K., TRIPLETT, C., AND BE...

Purification and Properties of an Extracellular Protease Produced by the Entomopathogenic Fungus Beauveria bassiana.

Beauveria bassiana GK2016 grown in a medium with gelatin as the sole carbon and nitrogen source produced an extracellular protease. The protease production was highest when the fungus was grown on a semiliquid medium and was purified about 18-fold, with a recovery of 21%. The protease molecular weight was estimated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis to be about 35,000....

THE DESIGN, MODELING AND EVALUATION OF POTENTIAL HIV PROTEASE INHIBITORS USING BLITZ, AN INTERACTIVE COMPUTER GRAPHICS WORKING TOOL

Several nonpeptide small molecules were designed as potential inhibitors of HIV protease and their structures were constructed by computer-aided molecular modeling and docked iwo the active site of HIV protease. Models of the complexes of inhibitors and the HIV protease were refined using nonbonded and H-bonding terms. The refined energy of selected complexes showed that the designed inhib...

DFT Studies and Topological Analyses of Electron Density on Acetophenone and Propiophenone Thiosemicarbazone Derivatives as Covalent Inhibitors of Falcipain-2, a Major Plasmodium Falciparum Cysteine Protease

Thiosemicarbazones (TSCs) possess significant antimalarial properties believed to be linked to the inhibition of major cysteine proteases, such as falcipain-2, in Plasmodium falciparum. However, the binding modes of TSCs to the active site of these enzymes are not clear. As a result of this, the nature of the bonding interactions between the active site of falcipain-2 and different derivatives ...

Evidence for the existence of a non-catalytic modifier site of peptide hydrolysis by the 20 S proteasome.

The 20 S proteasome is an endoprotease complex that preferentially cleaves peptides C-terminal of hydrophobic, basic, and acidic residues. Recently, we showed that these specific activities, classified as chymotrypsin-like, trypsin-like, and peptidylglutamyl peptide-hydrolyzing (PGPH) activity, are differently affected by Ritonavir, an inhibitor of human immunodeficiency virus-1 protease. Riton...
